Particles based on chitosan and fluorescein, prepared by encapsulation and chemical adsorption methods, were cross-linked using the soft cross-linking method. The degree of release of fluorescein particles on their surface was studied. The qualitative components of the obtained materials were determined by infrared spectroscopy. The loading and release degree of fluorescent particles encapsulated/adsorbed by polymer microspheres were quantitatively determined by spectrophotometry. To determine the release mechanism, the obtained fluorescence particle release data were analyzed according to zero-order and first-order, Higuchi and Korsmayr-Peppas models. Research has confirmed that the chitosan—fluorescein copolymer can precisely release drugs in specific physiological environments (such as the acidic environment around tumor cells, with a pH value of 5.0), enabling the drugs to target the tumor site. Moreover, its non-expanding diffusion mechanism can also be widely applied in the field of controlled-release preparations for long-term stable drug release. In the future, it can focus on developing biosensors based on chitosan materials, which can detect specific biological processes by monitoring the release kinetics of fluorescein.
