• eLiposomes represent a novel class of drug-delivery carriers comprising an O/W nanoemulsion within the aqueous core of liposomes. • Experimental bulk O/W nanoemulsions demonstrate long-term stability to aggregation and other degradation processes. • Langevin dynamics simulations reveal oil droplets self-assembling inside the confined space inside eLiposomes. • Depending on ζ-potentials, oil droplets form densely or sparsely packed layers on the inner surface of liposomes and these structures can be regarded as inverted colloidosomes.
