A polyherbal combination has been reported to exert synergistic effects between Phyllanthus niruri, Centella asiatica, Curcuma xanthorrhiza, and Curcuma longa. The pharmacological activity of these bioactive compounds is limited by their solubility and permeability; however, this limitation can be overcome through nanoemulsion (NE) technology, which significantly enhances absorption. The present study aimed to develop a self-nanoemulsion (S-NE) formulation of the polyherbal combination to improve the bioavailability of its active compounds. The NE components, oil, surfactant, and co-surfactant, were optimized using an integrated statistical approach, employing a categorical model factorial design for screening and a D-optimal design for formulation development. The optimized formulation was evaluated based on self-emulsification efficiency, droplet size and distribution, zeta potential, antioxidant activity, and drug loading capacity. Pharmacokinetic studies of the optimized formulation were also conducted. Results demonstrated that the modified oil reduced droplet size and polydispersity, the surfactant enhanced antioxidant activity, and the co-surfactant improved NE stability. The optimized formulation consisted of 29.08% Capmul PG 8 NF, 35.52% Kolliphor RH 40, and 35.41% Transcutol P. Pharmacokinetic evaluation revealed that the bioavailability of curcumin and phyllanthin, the principal compounds in the polyherbal combination, was nearly doubled across all parameters. This study highlights that S-NE formulations can effectively enhance the bioavailability and bioactivity of polyherbal combinations, offering a promising strategy for improving therapeutic outcomes.
