• Senolytic (D+Q) treatment alleviates cognitive decline and cellular senescence in the aging mouse brain. • Multi-omics reveals senolytics specifically downregulate the hippocampal cholesterol biosynthesis pathway. • The anti-aging effects of senolytics are localized to a reduction in lipid droplets within astrocytes. • Inhibiting astrocytic cholesterol synthesis is both necessary and sufficient for senolytic efficacy. • Genetic overexpression of HSD17b7 reverses the benefits of senolytics, establishing a causal mechanism.
