• PACS2 silencing inhibits Ang-II-triggered hypertrophy in cardiomyocytes. • PACS2 silencing promotes SIRT1 expression and activates Nrf2/HO-1 pathway. • PACS2 silencing improves cardiac hypertrophy by promoting SIRT1 expression. • HIC1 is a transcription regulatory factor of SIRT1. • PACS2 affects SIRT1 expression by binding to HIC1.
