Constitutively activated Signal Transducer and Activator of Transcription 3 drives human cancer development and progression and represents a validated drug target. Signal Transducer and Activator of Transcription 3-targeting modalities have been developed, although low potency, poor physicochemical properties, and toxicity have limited their clinical development. Protein-degrader strategies are overcoming potency challenges and accelerating the preclinical and clinical development of Signal Transducer and Activator of Transcription 3-targeting modalities. Outcomes for Signal Transducer and Activator of Transcription 3-targeting agents in clinical trials show some promise and suggest that more work remains to be done. Signal Transducer and Activator of Transcription 3-targeting agents, when combined with other drugs and delivered at lower doses, could offer opportunities to enhance clinical benefits while limiting overall toxicity.
