• Compressive strain significantly reduces glucose and lactate diffusivity in human TMJ discs. • Strain-induced decreases in porosity strongly correlate with impaired solute transport. • Sustained loading simulations predict localized nutrient depletion and reduced ATP synthesis. • Human TMJ discs exhibit species-specific strain–diffusion behavior compared to porcine models.
