• Monascus. rubiginosus (M. rbg) exhibited the greatest total tocopherol accumulation ability among the four Monascus spp. during coix seed fermentation. • The optimization strain (M. rbg-2 M) with a 2.98-fold increase in total tocopherol yield compared to the initial strain (M. rbg). • Supplementation with key regulation factors—L-Phenylalanine (L-Phe), L-Tyrosine (L-Tyr), and sodium dodecyl sulfate (SDS) significantly increased both total tocopherol enrichment and the diversity of tocopherol isomers. • The synergistic combination of strain optimization and fermentation regulation elevated total tocopherol in the M. rbg-2 M to 346.49 μg/g, over fifteen-fold higher than the M. rbg.
