Streptomyces kryoterrae sp. nov. and Streptomyces yamdrokensis sp. nov., two novel actinobacteria isolated from the Qinghai-Tibet Plateau

Two actinomycete strains, QTS 52T and QTS 137T, were isolated from permafrost and lakeshore soils of the Qinghai–Tibet Plateau, China. Phylogenetic analyses based on 16S rRNA gene sequences and whole-genome data confirmed their placement within the genus Streptomyces. Strain QTS 52T was most closely related to S. lacrimifluminis CGMCC 4.7272T (98.61% similarity), while QTS 137T was closest to S. pseudovenezuelae CGMCC 4.1754T (98.34%).The genomes of QTS 52T and QTS 137T were 10.07 Mb (70.13 mol% G + C) and 8.13 Mb (71.49 mol% G + C), respectively. Average nucleotide identity (ANI) and digital DNA–DNA hybridization (dDDH) values between the isolates and their closest relatives were below the accepted species thresholds (QTS 52T vs. S. lacrimifluminis: 92.47% ANI, 48.20% dDDH; QTS 137T vs. S. pseudovenezuelae: 80.82% ANI, 24.90% dDDH), supporting their status as novel species. Chemotaxonomic and phenotypic analyses further distinguished the strains: QTS 52T lacked aminolipid AL1 present in its relative, and QTS 137T did not produce H₂S and lacked diphosphatidylglycerol, unlike S. pseudovenezuelae. The major fatty acids were isoC16:0 and H-isoC16:1 for QTS 52T, and anteiso-C15:0, isoC16:0 and C16:1ω7c/C16:1ω6c for QTS 137T. Both strains produced laccase under inducing conditions, including at low temperatures, indicating retention of extracellular oxidative enzyme activity in these permafrost-derived isolates. Based on this polyphasic evidence, we propose Streptomyces kryoterrae sp. nov. (type strain QTS 52T = GDMCC 4.462T = JCM 37994T) and Streptomyces yamdrokensis sp. nov. (type strain QTS 137T = GDMCC 4.461T = JCM 37995T).
