• Picolinic acid substituents effect on Ag(I) and Zn(II) coordination. • Distinct speciation for silver(I) and zinc(II) 3-MePicH and 3-OHPicH complexes. • Structures confirmed by spectroscopic, thermal and single-crystal analysis. • Silver(I) complexes show enhanced antimicrobial and anticancer activity. • Strong DNA binding and topoisomerase II inhibition by Ag3-MePic.
