• Synthetic biology offers innovative solutions for TNBC, integrating engineered cells, gene-editing tools, and programmable microbes to overcome therapeutic limitations. • Logic-gated CAR-T cells improve tumor specificity and safety, representing a major advancement in cell-based immunotherapy for TNBC. • Engineered bacterial therapies demonstrate selective tumor targeting and significant antitumor activity in preclinical TNBC models. • iPSC-based platforms support patient-specific TNBC modeling and the generation of immune effectors like CAR-NK cells. • CRISPR/Cas9 technology enables precise editing of oncogenic drivers and resistance pathways, enhancing targeted TNBC therapy.
