• This review elucidates the research advancement on NLRP3 inflammasome from the aspect of structure, function and activation signaling. • It uncovers the inextricable role of NLRP3 inflammasome in AD pathogenesis from the preclinical and clinical evidence. • It highlights the capacities of different glia, cellular processes and signaling events that regulate the Aβ-induced NLRP3 inflammasome activation. • It overviews the existing anti-AD agents that inhibit the NLRP3 inflammasome activation.
