• Tumor cell-intrinsic POU4F1 contributes to the establishment of an immunosuppressive microenvironment in basal-like breast cancer. • POU4F1 promotes monocyte recruitment and immunosuppressive phenotype polarization of macrophages through NIK-P52/RelB-CCL2 axis. • Increased macrophage infiltration and immunosuppressive polarization inhibits the proliferation and effector functions of CD8+ T cells and NK cells. • Targeting POU4F1 inhibits tumor growth, restores antitumor immune response and potentiates immunotherapy.
