• TBX6 is aberrantly upregulated in colorectal cancer and predicts poor outcomes. • TBX6 promotes colorectal cancer cell proliferation, migration, invasion, and EMT. • TBX6 knockdown suppresses xenograft growth and enhances tumor cell apoptosis. • RNA-seq and PPI analyses identify CXCL14, NOTUM, ADRA2A, and MCHR1 as hubs. • TBX6 links CXCL14 repression to altered Wnt/β-catenin signaling in CRC.
