• Blocking the CLEC-2-PDPN axis prolongs skin allograft survival. • PDPN blockade reduces migratory cDC2 accumulation in lymph nodes. • Impaired cDC2 homing broadly suppresses T cell alloimmunity. • The therapy increases IL-10, fostering a tolerogenic state.
