• SiMyD88 was identified and characterized in the sea urchin Strongylocentrotus intermedius. • SiMyD88 expression was induced by LPS and poly(I:C) challenge. • SiMyD88 interacted with TLR proteins and was capable of self-association. • SiMyD88 interacted directly with IRAK. • SiMyD88 dose-dependently activated the IL-6, AP-1, and NF-κB reporter genes.
