• Based on metagenomic sequencing data, combined with WGCNA and machine learning, B. ovatus was identified as a potential key gut microbe that alleviates postmenopausal osteoporosis. • Through in vivo experiments and metagenomic sequencing, B. ovatus was verified to attenuate bone loss in OVX mice. • In vivo experiments confirmed that B. ovatus alleviates bone loss in OVX mice by producing IAA. • In vitro assays demonstrated that IAA interacts with AhR, upregulates NQO1 expression, suppresses intracellular ROS production, and consequently inhibits osteoclast differentiation.
