• A comprehensive overview of key regulated cell death (RCD) pathways is presented, detailing the molecular mechanisms of apoptosis, pyroptosis, ferroptosis, and necroptosis. • Intricate crosstalk and functional cooperativity among distinct RCD modalities are emphasized, culminating in the emerging concept of PANoptosis as a unified cell death machinery. • NINJ1 is highlighted as a universal executioner of plasma membrane rupture, representing a novel and promising therapeutic target for controlling the inflammatory outcome of lytic cell death.
