• 30 days, but not 15 days, of chronic heroin treatment could impair the learning and memory of mice in the water maze task • Prolonged heroin administration increased SIRT1- and HMGB1-positive microglia in the hippocampus. • Microglial SIRT1 knockdown worsened hippocampal apoptosis and cognitive impairment after 30 days of heroin treatment. • Microglial SIRT1 knockdown reduced the increase of HMGB1-positive microglia induced by chronic heroin treatment.
