• Phagocytic capacity against V. splendidus rather than S. aureus was specifically enhanced in the V. splendidus-primed oysters. • The cell-free haemolymph in the V. splendidus-primed oysters exhibited stronger antibacterial activities against V. splendidus. • The expression levels of phagocytosis-related and antimicrobial peptide genes in haemocytes upregulated in a microbe-specific manner. • The H3K4me1 and H3K9ac enriched in the haemocytes after stimulation with either V. splendidus or S. aureus.
