THEMIS: new insights into its evolution, structure, and function

Abstract

THEMIS (thymocyte-expressed molecule involved in selection) regulates signaling through the evolutionarily conserved cysteine-containing all-beta-in-THEMIS (CABIT) modules, unique structural motifs that emerged with the expansion of metazoan tyrosine phosphorylation networks. CABIT modules are fast evolving, suggestive of direct interaction with pathogens or pathogen-encoded proteins. Recent findings address long-standing controversies, providing evidence that THEMIS enhances T cell antigen receptor (TCR) signaling by inhibiting Src homology region-2 domain-containing tyrosine phosphatase-1 through its CABIT modules, but it also may have inhibitory effects on TCR signaling in certain contexts. THEMIS expression is dynamically regulated across T cell subsets, influencing homeostatic maintenance, effector functions, memory formation, and regulatory T cell activity. THEMIS serves as a modulator that balances protective immunity and the propensity for autoimmune or inflammatory diseases. Emerging evidence indicates that THEMIS operates as a regulator of the immune checkpoints B- and T- lymphocyte attenuator and PD-1, positioning THEMIS as a potential target for immunotherapy.
