• TRPC5 antagonism exerts mood-stabilizing effects in an ouabain-induced rat model of bipolar disorder. • The TRPC5 antagonist ML204 reverses behavioral changed induced by ouabain. • TRPC5 blockade restores BDNF–TrkB signaling and synaptic protein expression. • ML204 normalizes GSK3β signaling and reduces astrocytic activation in the hippocampus. • TRPC5 antagonism shows lithium-comparable efficacy on behavioral and molecular outcomes.
