• The bi-organellar regulator MNRR1 functions in the nucleus as a transcriptional activator by recruiting the co-activator p300 to the transcription factor RBPJκ. • MNRR1 levels are reduced in heteroplasmic MELAS cybrid cell lines. • The nuclear function of MNRR1 is sufficient to rescue mitochondrial deficit in a cellular model for MELAS. • Minimal MNRR1 peptides (TA-R1) can rescue transcriptional activation function of MNRR1.
