• Lipid overload decreases TRPM8 protein abundance and disrupts mitochondrial Ca2+ homeostasis in mouse ventricular myocytes. • L-menthol restores TRPM8 S-palmitoylation suppressed by lipid overload, thereby stabilizing TRPM8 protein expression. • L-menthol enhances TRPM8/GRP75/VDAC1 complex formation and attenuates lipid overload-induced mitochondrial Ca2+ dysregulation in mouse ventricular myocytes.
