• The fabricated MZSP NPs can efficiently generate ROS, disrupt ionic homeostasis and activate antitumor immunity. • MZSP NPs, rich in sulfur vacancies, release H2S in tumors to relieve hypoxia and boost sonodynamic ROS generation. • Zn2⁺ is rapidly released in response to the tumor microenvironment and ultrasound, enhancing the activation of Caspase-1. • ROS and Zn2⁺ synergize to disrupt mitochondria, inducing pyroptosis and boosting antitumor immunity.
