• PfTRAF6 was identified and characterized from yellow catfish. • PfTRAF6 expression was upregulated after Aeromonas hydrophila infection. • PfTRAF6 knockdown reduced LC3-II formation and P62 phosphorylation. • PfTRAF6 regulated autophagosome formation in monocytes/macrophages. • PfTRAF6 plays a key role in bacterial infection–induced autophagy in fish.
