Ultrasound-assisted synthesis of gold-platinum nanoparticles: Oxidase-like properties and nitrite detection in pickled food

Nanozymes, a class of nanomaterials with enzyme-like catalytic activities, face practical limitations due to nanoparticle aggregation during synthesis and insufficient long-term storage stability. To address this challenge, a silk fibroin peptide@gold-platinum (SFP@Au-Pt) nanozyme with high enzyme-like activity and storage stability was synthesized by ultrasound treatment in this study. SFP@Au-Pt by ultrasound synthesis exhibited a high affinity toward TMB (Km=0.048 mM) with a maximum reaction velocity (Vmax) of 0.067 mu M & sdot;s-1 . Following 15 days of storage at 4 degrees C, the relative retention rate of enzyme activity in the ultrasonic treatment group increased by 17.2% compared to the non-ultrasonic treatment group. Under acidic conditions, SFP@Au-Pt catalyzes the oxidation of colorless TMB into blue TMBox; in the presence of nitrite, a diazotization reaction between nitrite and TMBox results in a color transition from blue to yellow. This method demonstrates a broad linear response range (0.5-120 mu g/mL), a low detection limit (0.247 mu M), and excellent selectivity. This work not only provides a simple, environmentally friendly synthesis process for Au-Pt nanoparticles under mild reaction conditions but also expands rapid detection strategies for nitrites in food and offers valuable insights into the design of nanozymes with enhanced storage stability.
