• It was revealed that isosorbide dinitrate (ISDN) can be triggered to release nitric oxide under low-intensity ultrasound in the presence of vitamin B2. • Leveraging the property of ISDN, the engineered nanoplatform enables on-demand and spatiotemporally controlled release of nitric oxide. • When combined with ultrasound, the naonoplatform’s antibacterial and anti-biofilm capabilities were enhanced, as evidenced by a bacterial survival rate below 10% with the biofilm during the formation stage. • Superior preservation of alveolar bone and profound elimination of inflammation were achieved with this nanoplatform treatment in the rat periodontal disease model.
