• Proposed a non-invasive, label-free, microscale thermometry approach. • The temperature measurement can reach beyond 410 K and exhibit a sensitivity of 1.2 K. • Measuring the threshold temperature prior to bubble nucleation and cell temperature. • Measuring the thermal conductivity of transparent substrate. • Excellent compatibility with any standard optical microscope.
