• S-nitrosylation improved beef pH and WHC but decreased tenderness. • Label-free modificomics first identified 125% more S-nitrosylated proteins in beef. • S-nitrosylation modulates multiple metabolic processes, with ferroptosis as a novel pathway. • ACSL, CP, and TF are candidate biomarkers of S-nitrosylation-regulated ferroptosis. • Correlation analysis supports SNO-ferroptosis role in beef quality regulation.
