• A series of novel indole-chalcone derivatives were designed and synthesized. • SPR and pull-down experiments confirmed that ZSW could directly bind to HK2. • ZSW significantly inhibits HK2 enzyme activity and LPS-induced inflammatory. • ZSW could modulate macrophage glycolysis and inflammation for IBD treatment.
