• 24 coumarin-quinoline hybrids were designed as multi-target-directed ligands for Alzheimer's multifaceted nature. • Lead compound B8 inhibited key targets (Aβ aggregation, cholinesterase, BACE1) and crossed the BBB. • B8 improved cognition in AD mice by reducing oxidative stress, inflammation, and Aβ deposition. • The hybrid strategy and compound B8 demonstrate significant promise for novel AD therapy development.
