• This rise in antimicrobial-resistant pathogens has led to significant morbidity and mortality. • Natural products exhibit a remarkable variety of scaffolds, and their contributions have been crucial in drug discovery. • Polyenes are lipophilic or amphipathic compounds that readily permeate the membranes of pathogens, inhibiting their growth. • Polyene-based compounds such as amphotericin, nystatin, and natamycin are used to treat fungal diseases, with the lowest likelihood of developing resistance. • The natural origin of polyenes underscores the ongoing need to enhance antibiotic drug discovery efforts.
