• HEWL is a natural antimicrobial protein whose defined structure and amphiphilic character confer remarkable structural adaptability. • Physicochemical mechanisms governing HEWL self-assembly are systematically summarized. • Diverse HEWL-based architectures-amyloid fibrils/nanotubes, microbubbles, hydrogels, micro/nanoparticles, emulsions, and films-are reviewed. • HEWL serves as a sustainable building block for multifunctional innovation in food formulation, active compound delivery, preservation, and biosensing. • Key challenges include structural heterogeneity, allergenicity, and biocompatibility.
