• Analytical hydrophobic interaction chromatography combined with LC-MS to evaluate hydrophobicity of CHO cell proteins • GRAVY number provides very poor prediction of HCP hydrophobicity • Calculations using 3D protein structure calculated by AlphaFold in better agreement with measured hydrophobicity • Results demonstrate effectiveness of analytical HIC coupled with LC-MS to explore hydrophobicity of complex mixtures
