• Pseudomonas fluorescens mechanosensitive-channel mutants were constructed to investigate ultrasound-induced stress responses. • MscL as the primary ultrasound mechanosensor rapidly relieved membrane tension through osmotic regulation. • Acoustic-pressure activation thresholds for MscL and MscS were 289.45 and 334.21 kPa, respectively. • MscL and MscS-deleted mutants aggravated ultrasound-induced membrane potential disruption, ATP depletion, intracellular component leakage, and oxidative stress. • MscL governed early responses of ultrasound while MscS modulates sustained and amplified signaling.
