• Polyhedral alkanes were used as linkers for the first time in PROTACs targeting ALK. • HAPOD trail was firstly used to assess the feasibility of PROTACs targeting ALK designing. • Five new PROTACs targeting ALK were synthesized via established organic synthesis routes. • Cell proliferation inhibition assay and Western blotting assay were used in vitro. • Molecular Dynamic Simulation analysis was used in silico.
