• Efficient CALB-catalyzed synthesis of MOGGs via two-step esterification. • Optimized conditions produced 62.7% MOGG yield with controlled DOGG formation. • Qualitative docking supported comparison of substrate accommodation in CALB. • Medium-chain fatty acids showed superior acylation efficiency toward GG. • Sequential solvent-free synthesis without intermediate GG purification.
