• IRF4-C99R mutations are recurrent in PMBCL and lead to a differentiation block, elevated TARC expression, and regulatory T-cell chemotaxis. • IRF4 mutation–associated upregulation of EPHB1 leads to lymphoma organotropism favoring thymic involvement.
